If you were Registered and logged in, you could reply and use other advanced thread options Start with a known clean machine (must not skip this !!)
Turn on the firewall
Take control over the passwords of all accounts, giving them
new, long, strong pass strings
Make sure you know of all accounts
start / run cmd
and then enter
net localgroup administrators
net localgroup users
Take steps to make sure the machine remains clean and updated
(anti-malware kept updates, Microsoft Update allowed to keep
your machine healthy)
There are more that may be done, but just those would be
sufficient for most home users

Are you talking about stopping someone that you believe has already accessed
your machine, or are you talking in the abstract, about prevention?
The answer to your main question is really going to depend on the answer to
that.
The simplest way to prevent remote access is to disconnect your machine from
the Internet when you aren't using it.
